PNC-27
also known as: p53 N-terminal peptide
Half-life: Unknown
About
PNC-27 is a synthetic peptide incorporating the MDM-2-binding domain of p53 fused to a membrane-targeting sequence. In vitro and animal studies suggest it selectively kills cancer cells by inducing membrane lysis in cells that overexpress HDM-2 (the human version of MDM-2), while sparing normal cells. Research compound only.
Mechanism of Action
PNC-27 contains the p53 residues 12-26 that bind MDM-2. In cancer cells with elevated HDM-2 (which sequesters p53), the peptide is believed to insert into the cell membrane and form pores, leading to selective cancer cell death through membranolysis.
Primary Benefits
- In vitro anti-cancer activity (breast, pancreatic, melanoma cell lines)
- Selective cancer cell targeting
- Research-stage oncology compound
Side Effects & Considerations
- Very limited human safety data
- Potential off-target membrane disruption
- Research use only
Research Notes
Sarafraz-Yazdi et al. (Oncotarget) have published in vitro data showing selective cytotoxicity against cancer cell lines. No clinical trials in humans.
