ACE-031 is a recombinant fusion protein that acts as a myostatin/activin type IIB receptor decoy, blocking inhibitory signals on muscle growth. It demonstrated substantial lean mass gains in clinical trials for Duchenne muscular dystrophy before development was halted due to vascular side effects at high doses.
Mechanism of Action
Binds and sequesters myostatin and related TGF-β ligands (activin A/B, GDF-11) before they reach the ActRIIB receptor, removing the brake on skeletal muscle protein synthesis.
Phase 2 trial (NCT01099371) in Duchenne MD showed lean mass increases of 2.7 kg vs placebo over 3 months at 1 mg/kg. Development paused due to vascular side effects at higher doses.
IGF-LR3 drives muscle protein synthesis downstream while ACE-031 removes the myostatin brake — powerful combination for hypertrophy.
Both block myostatin through different mechanisms; stack only at conservative doses due to additive side-effect risk.
Overlapping myostatin inhibition — combining at full doses is redundant and increases bleeding risk.
Use BPC-157 during breaks to support connective tissue that rapid muscle growth may stress.
Storage
Refrigerate at 2–8°C (36–46°F). Do not freeze.
Reconstitution
Typically supplied pre-dissolved; if lyophilized, reconstitute with sterile water for injection.
