also known as: Bremelanotide, Vyleesi, PL-6983
Half-life: ~2–3 hours
PT-141 (Bremelanotide) is a synthetic melanocortin receptor agonist derived from MT-2 by removing the cyclic lactam bridge. It is FDA-approved (as Vyleesi) for hypoactive sexual desire disorder (HSDD) in premenopausal women, and is widely used off-label for sexual dysfunction in both sexes. Unlike PDE5 inhibitors, it acts centrally on the brain to create desire, not just physical arousal.
Mechanism of Action
Activates melanocortin MC3R and MC4R receptors in the hypothalamus, triggering dopaminergic and oxytocergic pathways that produce sexual desire, arousal, and motivation. Works independently of the vascular system — effective even when PDE5 inhibitors fail.
FDA-approved June 2019 as Vyleesi (bremelanotide) for HSDD. Palatin Technologies phase 3 trial showed significant improvement in satisfying sexual events and desire vs placebo.
PT-141 drives physical arousal and desire; Oxytocin enhances emotional bonding and connection — complementary sexual enhancement.
MT-2 adds tanning and additional MC receptor stimulation — but combining increases nausea and side effects significantly.
MT-2 is the parent compound of PT-141 and acts on the same receptors — avoid combining.
