also known as: KL1, Klotho fragment, αKlotho peptide
Half-life: Unknown for injected fragment
Klotho Derived Peptide 1 (KL1) is a fragment of the α-Klotho protein, an aging-suppressor protein encoded by the KL gene. Blood levels of α-Klotho decline with age and are inversely correlated with cardiovascular disease, cognitive decline, and mortality. KL1 has shown cognitive improvements in aging mice at very low doses.
Mechanism of Action
The KL1 domain of α-Klotho modulates FGF23/FGFR signaling, improves synaptic plasticity via GluN2B-containing NMDA receptors, and exerts antioxidant and anti-inflammatory effects in the brain and kidneys.
Dubal et al. (Cell, 2014) showed systemic klotho elevation via gene injection improved cognitive function in young and aging mice at 10% elevation. Leon et al. (2023) demonstrated KL1 fragment improves working memory in aged mice.
