also known as: PNB-0408, N-hexanoic-Tyr-Ile-(6) aminohexanoic amide
Half-life: Long — estimated days due to tight receptor binding
Dihexa is a potent nootropic peptide derived from angiotensin IV. It is one of the most powerful known cognitive enhancers in animal models, reported to be approximately 7 orders of magnitude more potent than BDNF at promoting synapse formation. It acts through the HGF/Met pathway to promote neuroplasticity.
Mechanism of Action
Potentiates HGF (hepatocyte growth factor) binding to its receptor c-Met, dramatically upregulating synaptogenesis, neuronal survival, and memory consolidation pathways in the hippocampus and cortex.
Published in PNAS by Joseph Harding et al. (2013). Animal models show reversal of memory deficits from hippocampal damage. No human clinical trials completed.
Semax raises BDNF acutely; Dihexa potentiates HGF/Met for longer-term synaptogenesis — different but complementary neurotrophin pathways.
Selank provides anxiolytic and memory enhancement via GABA/serotonin modulation alongside Dihexa's structural synaptic work.
NAD+ restores neuronal energy metabolism, supporting the energy-demanding synapse formation Dihexa promotes.
