also known as: Kisspeptin-10, Kisspeptin-54, Metastin, KP-10, KP-54
Half-life: KP-10: ~3–5 minutes IV; ~20–40 minutes SubQ. KP-54: ~30 minutes IV.
Kisspeptin is a naturally occurring neuropeptide encoded by the KISS1 gene that acts as the master regulator of the hypothalamic-pituitary-gonadal (HPG) axis. By binding to GPR54 receptors in the hypothalamus it triggers a powerful pulse of GnRH, which in turn drives LH and FSH release, stimulating endogenous testosterone and estradiol production. It is used clinically to restore suppressed HPG axis function and is being studied extensively for male and female hypogonadism, IVF protocols, and as an alternative to exogenous LH in fertility treatment.
Mechanism of Action
Binds GPR54 (KISS1R) receptors on GnRH neurons in the hypothalamus. This removes tonic inhibition and drives pulsatile GnRH secretion, producing downstream LH and FSH surges from the pituitary and subsequent gonadal steroidogenesis (testosterone in men, estradiol/progesterone in women). Kisspeptin is also expressed in peripheral tissues including the placenta, liver, and gonads.
Dhillo et al. (JCEM 2005) — first human study showing IV KP-54 raises LH. Jayasena et al. (Clinical Endocrinology) demonstrated SubQ KP-54 sustained LH and testosterone elevation. Multiple clinical trials ongoing for male infertility (hypogonadotropic hypogonadism) and as an IVF ovulation trigger replacing hCG.
Kisspeptin drives GnRH release upstream while gonadorelin provides direct GnRH stimulation — layered HPG axis support for PCT.
PT-141 acts centrally on melanocortin receptors for direct sexual arousal; Kisspeptin raises testosterone to amplify baseline libido — complementary mechanisms.
Both ultimately raise LH/FSH via the HPG axis; using both simultaneously may cause overstimulation — use sequentially or at reduced doses.
BPC-157 supports vascular and tissue health during hormonal recovery cycles.
Adding a GH secretagogue alongside HPG axis restoration addresses both GH and testosterone axes for comprehensive recovery.
Storage
Lyophilized: store at -20°C (-4°F) long-term, 2–8°C (36–46°F) after reconstitution. Use within 2–4 weeks once reconstituted.
Reconstitution
Reconstitute with bacteriostatic water. Use 1–2 ml per vial. Store reconstituted peptide at 2–8°C (36–46°F).
