also known as: FOXO4-p53 DRI peptide, Senolytic peptide
Half-life: D-peptides: resistant to proteases, likely hours to days
FOX04-DRI is a D-retro-inverso peptide that disrupts the interaction between FOXO4 and p53 in senescent cells, allowing p53 to trigger apoptosis. It is a highly targeted senolytic — selectively eliminating senescent ("zombie") cells that accumulate with aging and drive inflammation and tissue dysfunction.
Mechanism of Action
FOXO4 protein in senescent cells sequesters p53 in the nucleus to prevent apoptosis. FOX04-DRI peptide competes with FOXO4, freeing p53 to initiate programmed cell death specifically in senescent cells while sparing normal cells.
Published in Cell by Peter de Keizer et al. (2017) showing selective senescent cell killing and restoration of aging biomarkers in old mice. No human trials completed.
Epithalon prevents new telomere shortening (senescence prevention); FOX04-DRI clears existing senescent cells — complementary anti-aging attack.
After FOX04-DRI clears senescent cells, GHK-Cu promotes regeneration of the cleared tissue.
Use BPC-157 after the senolytic week to support tissue repair in cleared areas.
