also known as: Mitochondrial open reading frame peptide, Mitokine
Half-life: ~4 hours
MOTS-C is a 16-amino acid peptide encoded within the mitochondrial genome (12S rRNA gene). It functions as a "mitokine" — a hormone-like signaling molecule produced by mitochondria that regulates systemic metabolism. Blood levels decline with age, and it is strongly implicated in age-related metabolic decline.
Mechanism of Action
Activates AMPK and inhibits the folate cycle/AICAR pathway, increasing glucose uptake and utilization. Translocates to the nucleus under metabolic stress to regulate nuclear gene expression. Improves insulin sensitivity and mitochondrial biogenesis.
Kim et al. (Cell Metabolism, 2015) identified MOTS-C as a mitochondrial-derived peptide that improves insulin sensitivity and exercise capacity. Janssens et al. showed MOTS-C declines in aging and its supplementation extends healthy lifespan in mice.
NAD+ and MOTS-C both support mitochondrial function through complementary mechanisms — the top mitochondrial duo.
SS-31 protects the inner mitochondrial membrane; MOTS-C signals systemic metabolic adaptation — comprehensive mitochondrial support.
5-Amino-1MQ raises NAD+ which fuels MOTS-C's AMPK activation pathway.
